Johnathan Hankins was born on March 30, 1992, in Dearborn Heights, Michigan, and played at Southeastern High School in Detroit. He was a first-team All-Big Ten defensive tackle at Ohio State in 2012. The New York Giants drafted him in the second round, 49th overall, in 2013. He played 11 games without a start as a rookie, and Madden NFL 25 rated him 70.

Hankins started all 16 games in 2014 and recorded a career-high seven sacks. Madden NFL 16 rated him 86 for the season that followed, which ended after nine games. He returned to start all 16 games in 2016, and Madden NFL 17 rated him 81.

He signed a three-year contract with Indianapolis in April 2017 and started 15 games before the Colts released him in March 2018. He then spent parts of five seasons with the Raiders, who traded him to Dallas in October 2022. Madden NFL 24 rated him 74 during the 2023 season, when he started 14 games for the Cowboys. He signed with Seattle in 2024 and played 17 games, then missed the 2025 season with a back injury. Hankins retired in July 2026 with 433 tackles and 18.5 sacks in 169 games across 12 seasons.
